Unbalanced low coherence interference microscopy

چکیده

Low coherence interference microscopy (LCIM) provides high spatial phase sensitivity, i.e., speckle free and coherent noise quantitative images of the test specimens. Due to low temporal (TC) length light source, LCIM requires precise adjustment optical path difference (OPD) between object reference arm, which is only a few micrometers. Consequently, previously demonstrated systems are implemented with use identical objective lenses in both arms also known as balanced interferometric configuration. The lens hinders numerical aperture swift change during imaging. In present work, non-identical arm called unbalanced A range 10 × /0.25NA, 20 /0.45NA 60 /1.2NA employed system while keeping single /0.25NA arm. To resolve challenges associated configuration, advanced iterative algorithm (AIA) principal component analysis (PCA) algorithms integrated recover quadratic error capabilities proposed method exhibited on various specimens like USAF resolution, step-like for biological cells, HeLa cells. approach enables scalable magnification resolution by simply rotating imaging turret without need changing
